    • can’t grow all things in pure culture (maybe 10% of bacteria found in humans can be grown this way)

    • no suitable animal model

    • organism is usually gone before symptoms develop

    • many organisms can produce the same syndrome

    • even nonpathogens can cause disease in the immunocompromised host
