Limitations of Koch’s postulates
can’t grow all things in pure culture (maybe 10% of bacteria found in humans can be grown this way)
no suitable animal model
organism is usually gone before symptoms develop
many organisms can produce the same syndrome
even nonpathogens can cause disease in the immunocompromised host
Molecular Koch’s postulates: how do you figure out if a gene leads to disease?
cloning
mutagenesis
Idealized approach to analysis of new bacterial virulence factors
in vitro ex vivo in vivo target for therapy?
