Pyrazinamide

  • First line anti tubercular agent
  • Active against intracellular bacilli in acidic pH


Mechanism of Action

  • Like Isoniazid it inhibits mycolic acid biosynthesis but through a different mechanism.
  • It is tuberculocidal


Pharmacokinetics

  • Orally administered and well absorbed from the gastrointestinal tract
  • Widely distributed across all the body cavities
  • Attains very high concentrations in the CSF and is very important agent in Tubercular meningitis.
  • Metabolised in the liver and excreted in urine.


Adverse Effects

  • Hepatitis
  • Interferes with the excretion of Uric acid - Hyperuricemia
    • May precipitate acute attacks of gout
