Some key steps in TLR signaling via NF-κB

  • TLR has extracellular domain

  • it binds to microbial-specific antigens such as LPS

  • it recruits an adaptor protein, one of which is MyD88

  • this starts a signaling cascade. at the end of the cascade, Iκk phosphorylates IκB

  • phosphorylated IκB is degraded, releasing NF-κB to migrate into the nucleus and regulate gene transcription
