Chemotherapy drugs and the cell cycle

  • antimetabolites and antifolates, as well as hydroxyurea, work in S phase because they deprive the cell of things necessary for DNA synthesis

  • microtubule inhibitors inhibit the cell in mitosis

  • phase-nonspecific drugs include alkylators, platinum agents, topo II inhibitors, bleomycin

Organizing these drugs another way

  • about half of our drugs work by impairing DNA synthesis

  • the other half of the drugs work outside of DNA synthesis. these drugs (platinum agents, alkylators, topo inhibitors) damage DNA

  • finally, the last drugs inhibit proteins (microtubule inhibitors, -ibs, and -abs)
